    "Born out of Crisis": an analysis of moorland management agreements on Exmoor; final report

    The designation of Exmoor National Park in 1954 recognised the importance of the open character of much of the area and, in particular, its moorland to that natural heritage of the UK. However, the moorland that the park was charged with maintaining and enhancing was soon under threat and the following years saw extensive moorland loss and the fragmentation of remaining moorland (Lowe et al, 1986). The ensuing moorland conflict and debate eventually lead Exmoor NPA, in collaboration with farmers, land owners, the CLA and NFU to pioneer a new system of moorland management agreements. Indeed, for a short time, Exmoor was the only location operating wholly voluntary management agreements (Brotherton, 1990). Only two agreements now remain and in the twenty-five years since their inception much has changed. Once castigated for their "theft of the countryside" (Shoard, 1980), many farmers now work in partnership with statutory and non-statutory organisations as 'stewards' of the countryside.     Regulation of Alpha- and Beta-actin Isoforms in the Contracting A7r5 Smooth Muscle Cell

    Two isoforms of actin have been found to be present in A7r5 smooth muscle cells, α- and β-actin. This body of work sets out to examine the different regulatory factors of the two actin isoforms during smooth muscle contraction. The response of the actin isoforms to phorbol 12, 13-dibutyrate (PDBu) is markedly different. α-Actin remodels to podosomes around the periphery of the cell, while β-actin merely shortens. One protein involved in smooth muscle contraction induced by PDBu is protein kinase C (PKC). Two inhibitors of PKC, staurosporine and bisindolymaleimide, were used prior to PDBu stimulation and after PDBu stimulation to observe the effects of PKC on α- and β-actin stress fiber structure and remodeling. α-Actin showed a decrease in podosome formation when the inhibitors were added prior to or after PDBu stimulation. β-actin demonstrated a loss in stress fiber structure in response to PKC inhibitors. PKC appears to regulate α- and β- actin differently and could play a role in the differential remodeling seen by these two actin isoforms. Myosin light chain kinase (MLCK) is another enzyme involved in smooth muscle contraction. Even though it does not play a major role in PDBu induced contraction of smooth muscle, it has been found to interact with actin at two different actin binding sites, allowing crosslinking of actin filaments. Inhibition of the kinase domain of MLCK does not seem to have an effect on the crosslinking abilities of MLCK. These two key enzymes of smooth muscle contraction appear to not only be able to initiate smooth muscle contraction, but also regulate reorganization of the two actin isoforms during smooth muscle contraction

    Anaplastic Lymphoma Kinase (ALK) is a receptor tyrosine kinase aberrantly expressed in a variety of tumor types, most notably in Anaplastic Large Cell Lymphoma (ALCL) where a chromosomal translocation generates the oncogenic fusion protein, Nucleophosmin-ALK (NPM-ALK). Whilst much is known regarding the mechanism of transformation by NPM-ALK, the existence of cellular defence pathways to prevent this pathological process has not been investigated. Employing the highly tractable primary murine embryonic fibroblast (MEF) system we show that cellular transformation is not an inevitable consequence of NPM-ALK activity but is combated by p53 and Rb. Activation of p53 and/or Rb by NPM-ALK triggers a potent proliferative block with features reminiscent of senescence. While loss of p53 alone is sufficient to circumvent NPM-ALK-induced senescence and permit cellular transformation, sole loss of Rb permits continued proliferation but not transformation due to p53-imposed restraints. Furthermore, NPM-ALK attenuates p53 activity in an Rb and MDM2 dependent manner but this activity is not sufficient to bypass senescence. These data indicate that senescence may constitute an effective barrier to ALK-induced malignancies that ultimately must be overcome for tumor development

    Supporting interactive system testing with interaction sequences

    Despite extensive research into the modelling and testing of interactive systems, existing strategies do not adequately cover all parts of an interactive system. These existing strategies model and test either the functional or interactive components of an interactive system separately, however, issues may arise where these components intersect. Therefore, further investigation into the modelling and testing of this intersection is required. Interaction sequences are a series of steps a user can take to complete a specific task or to arbitrarily explore an interactive system. In this research interaction sequences are used as an abstraction of the interactive system to inform a model-based testing approach using lightweight formal methods. Interaction sequences provide an abstract view of the point at which the functional and interactive components intersect, and as a result also provide a good starting point for investigation into the modelling and testing of this area. Interaction sequences are applicable to all types of interactive systems irrespective of the type of interaction, therefore modelling and testing approaches using this abstraction are also applicable to all types of interactive systems. In this thesis the findings of our investigation into modelling and testing using interaction sequences are presented. We describe formalisation of interaction sequences and modelling of these sequences using Finite State Automata (FSA). We introduce the self-containment property and show how this is used to control the size and state space of FSA. We demonstrate simulating interaction sequences and discuss how these models can be applied within both model checking and testing techniques. Lastly, we present a new approach for generating tests from interaction sequences and their associated models

    Effects of nebulised magnesium sulphate on inflammation and function of the guinea-pig airway

    Magnesium sulphate is a potential treatment for acute severe asthma. However, the mechanisms and dose-response relationships are poorly understood. The first objective of this study was to examine whether inhaled magnesium sulphate exerts bronchodilator activity measured as bronchoprotection against histamine-induced bronchoconstriction in conscious guinea-pigs alone and combined with salbutamol. Secondly, we examined whether inhaled magnesium sulphate inhibits airways inflammation and function in models of neutrophilic and eosinophilic lung inflammation induced, respectively, by inhaled lipopolysaccharide or the inhaled antigen, ovalbumin (OVA). Airway function was measured in conscious guinea-pigs as specific airway conductance (sGaw) by whole-body plethysmography. Anti-inflammatory activity was measured against lung inflammatory cell influx induced by OVA inhalation in OVA-sensitised animals or by lipopolysaccharide (LPS) exposure of non-sensitised animals. Airway function (sGaw) was measured over 24 h after OVA exposure. Airway hyperresponsiveness to inhaled histamine and inflammatory cells in bronchoalveolar lavage fluid were recorded 24 h after OVA or LPS challenge. Histamine-induced bronchoconstriction was inhibited by inhaled magnesium sulphate or salbutamol alone and in combination, they produced synergistic bronchoprotection. LPS-induced neutrophil influx was inhibited by 6 days pretreatment with magnesium sulphate. Early and late asthmatic responses in OVA sensitised and challenged animals were attenuated by magnesium sulphate. Lung inflammatory cells were increased by OVA, macrophages being significantly reduced by magnesium sulphate. Nebulised magnesium sulphate protects against histamine-induced bronchoconstriction in conscious guinea-pigs and exerts anti-inflammatory activity against pulmonary inflammation induced by allergen (OVA) or LPS. These properties of magnesium sulphate explain its beneficial actions in acute asthma

    The Changing Role of Ancillary Health Care Service Providers: An Evaluation of Health Diagnostic Laboratory, Inc.

    In an effort to reduce cost and improve quality, health care payers have enacted a number of incentives to motivate providers to focus their efforts on achieving better clinical outcomes and reducing the prevalence and progression of disease. In response to these incentives, providers are entering into new arrangements such as accountable care organizations and patient-centered medical homes to redesign delivery processes and achieve quality and cost objectives. This article reports the results of a study designed to evaluate the impact on cost and quality of care resulting from services provided by Health Diagnostic Laboratory, Inc., a clinical laboratory with a comprehensive care model. The results show that patients who utilized these laboratory services experienced lower total cost of care (23% reduction, P \u3c 0.01) and improved lipid profiles during the follow-up period. Total cost reductions were related to cost reductions found in both inpatient and ambulatory care. These findings suggest that accountable care organizations, patient-centered medical homes, and other groups entering shared savings initiatives should consider the potential role ancillary service providers with comprehensive care models can play in the delivery of integrated care.     Modeling Multi-Wavelength Stellar Astrometry. II. Determining Absolute Inclinations, Gravity Darkening Coefficients, and Spot Parameters of Single Stars with SIM Lite

    We present a novel technique to determine the absolute inclination of single stars using multi-wavelength sub-milliarcsecond astrometry. The technique exploits the effect of gravity darkening, which causes a wavelength-dependent astrometric displacement parallel to a star's projected rotation axis. We find this effect is clearly detectable using SIM Lite for various giant stars and rapid rotators, and present detailed models for multiple systems using the REFLUX code. We also explore the multi-wavelength astrometric reflex motion induced by spots on single stars. We find that it should be possible to determine spot size, relative temperature, and some positional information for both giant and nearby main-sequence stars utilizing multi-wavelength SIM Lite data. This data will be extremely useful in stellar and exoplanet astrophysics, as well as supporting the primary SIM Lite mission through proper multi-wavelength calibration of the giant star astrometric reference frame, and reduction of noise introduced by starspots when searching for extrasolar planets.Comment: 8 pages, 7 figures, 4 tables.
